Actin heterogeneity in the Fiocco of the Casertana black pig AAGT [Ancient Autochthonous Genetic Type; Campania]

The characterisation of Fiocco Sannita, seasoned meat product obtained from the ancient autochthonous pig Casertana, was carried out by means of proteomics approach after fractionating meat proteins according to their solubility. Two-dimensional electrophoresis maps of sarcoplasmatic and myofibrillar protein fraction soluble in low ionic force and denaturing and reducing buffer, respectively, evidenced a complex pattern. This complexity is due, in addition to the main sarcoplasmatic and myofibrillar proteins, also to their isoforms and fragments originated from proteolysis which occurs during meat maturation. The research was carried out on 12 Fiocco Sannita samples and proteins were identified by mass spectrometry MALDI-TOF. La caratterizzazione del Fiocco Sannita, prodotto stagionato ottenuto dal suino autoctono antico Casertana, è stata effettuata mediante approccio proteomico, previo frazionamento delle proteine della carne in base alla loro solubilità. Le mappe bidimensionali delle frazioni sarcoplasmatica solubile in tampone a bassa forza ionica e miofibrillare solubile in tampone denaturante e riducente hanno evidenziato una notevole complessità. Tale complessità è dovuta, oltre che alle principali proteine sarcoplasmatiche e miofibrillari, anche ad alcune loro isoforme e frammenti derivanti dalla proteolisi che avviene durante il processo di maturazione della carne. La ricerca è stata condotta su 12 Fiocco Sannita e le proteine sono state identificate con l'ausilio della spettrometria di massa MALDI-TOF. In questa indagine sono state rivelate e identificate tre forme di actina mai descritte finora per il muscolo suino
