Genotyping of S-RNase gene in sweet cherry [Prunus avium] cultivars by PCR and crossing

1. A simpler method of PCR-based typing system of sweet cherry (Prunus avium L.) cultivars was developed for six S-alleles (Ssup(1), Ssup(2), Ssup(3), Ssup(4), Ssup(6), Ssup(9)) based on the DNA sequence data obtained from the cDNAs encoding S-RNases (Tao et al., 1999). 2. In the present study, we investigated S-genotypes of sweet cherry cultivars in the Yamanashi Fruit Tree Experiment Station and the elucidation of S-genotypes was confirmed by the references and the pollination test using 'Saori', 'Rainier' and 'Taishounishiki'. The results of the present study may contribute in selecting the pollinizer in the sweet cherry industry.
