Two hundred and sixteen leaf samples of potato varieties viz. Cardinal, Diamont and Desiree were analyzed and compared to know the status of the material being produced by rapid multiplication sources through Enzyme Linked Immunosorbent Assay (ELISA) against potato Virus X (PVX), Potato Virus Y (PVY) and Potato Virus S (PVS) in Ayub Agricultural Research Institute Faisalabad during 2004-05. The samples were collected from three sources i.e. Punjab Seed Corporation (PSC), Ayub Agricultural Research Institute, (AARI) and Nuclear Institute of Biotechnology and Genetic Engineering (NIBGE). Out of seventy two samples of each variety from three sources, two samples of Cardinal, three samples of Diamont and four samples of Desiree were infected with PVX. The maximum (2.88%) PVX recovery was found in the samples of PSC as compared to NIBGE (2.16%) and AARI (1.44%). Three samples of each of Cardinal and Diamont, five samples of Desiree were infected with PVY. The maximum PVY was found in the samples of PSC (5.04%), followed by NIBGE (2.16%) and ARRI (0.72%). Three samples of Cardinal, four of Diamont and seven samples of Desiree were infected with PVS from three sources. The maximum PVS recovery was found in the samples of PSC (5.46%) followed by NIBGE (2.88%) and AARI (1.44%).
