[Study of genome of cultivated plants and their congeners in apply to the genetic breeding theory: study of desemfins spectra in phytopathogene-resistant and susceptible wheat species]

Defensins were isolated and sequenced from the seeds of high resistant species Triticum kiharae, susceptible varieties of common wheat Khakasskaya and Rodina, as well as T. monococcum (supposed as a genome A donor), Aegilops speltoides (genome B), Ae. squarrosa (genome D). Thirteen defensins from T. kiharae were divided to 3 groups by homology of N-terminal sequences. Group 1 covered 8 defensins named D-defensins. Their similarity to the amino acid sequences the wheat defensin TADI induced by cold adaptation has been found. Group 2 consisted of 3 peptides, 2 of them had been detected before in seeds of T. durum. In group 3 there were 2 new peptides homologous to barley defensins. High conservatism of the primary structure of D-defensins is observed. Genomic localization of their genes has been found. Defensins D1, D4 and D5 are controlled by genome A, D3 and D6 – by genome D, and D6.1 and D3.1 – by genome B. The varieties Khakasskaya and Rodina did not differed in the composition of desenfins from T. kiharae in spite of the differences of genomic composition (ABD in T. aestivum and AGD in T. kiharae). It is concluded about the absence of a direct relationship between the composition of D-desenfins and the level of resistance in compatible plant-pathogen combinations.
