The objective of this study was to examine the organization of the actin cytoskeleton in human embryonic kidney cells (HEK 293 cells) that had been stably transfected with a thyrotropin-releasing hormone (TRH) receptor. The emphasis was on its rearrangement induced by the activation of G-protein coupled receptors (GPCR) exemplified by the TRH receptor. Morphometric cell analysis was performed at the light microscope level. Distribution of fluorescently-labelled actin filaments was monitored by confocal microscopy. Agonist stimulation of cells expressing the TRH receptor resulted in an increase in their surface area and the reorganization of their actin network. This agonist-induced effect can be abolished by the concomitant treatment of the cells with cytochalasin D. These results demonstrate that an agonist-induced activation of the TRH receptor, a member of the GPCR family, causes a change in a cell's morphology that probably correlates with the rearrangement of the actin cytoskeleton.
