Isolation and expression of chitinase and glucanase in flax zygotic embryos

Here we present isolation of a chitinase gene sequence from flax. On mRNA from embryogenic and non-embryogenic flax calli a RT-PCR and PCR were performed using both degenerated and specific primers. The partial, 245 bp long sequence reveals at both nucleotide and protein levels homology to chitinases from other plant species such as chestnut, soybean and apple. In addition, a DNA fragment of glucanase, an another glucanhydrolase, has been isolated from flax. The sequence of 628 bp shows identity with other plant glucanases, e. g. bean and wheat. Both sequences isolated are reported in the GenBank, access. No. AY847514, AY780204. Our preliminary expression studies of these two genes in flax zygotic embryos could indicate their embryogenic potential.
