The effect of different cytokinins (BA, KIN, 2-iP, TDZ, ZEA) on the initiation of somatic embryogenesis in hybrid firs Abies alba x A. cephalonica (cell lines AC 2, AC 79) and Abies alba x A. numidica (AN 72) has been tested. Cotyledons dissected from emblings have been used as explants. In cell lines AC 2 and AN 72 initiation occurred by all the tested cytokinins. In cell line AC 79 ZEA and 2-iP were unsuccessful giving no embryogenic tissue. Initation frequencies reached maximum in the presence of BA (5.55 to 14.87%). The somatic embryos formed directly on the explants without previous callus phase. The process of somatic embryo development begins with cell division in epidermal and subepidermal cell layers and leads to the formation of spherically arranged clusters of meristematic cells. Polarisation of spherical structures resulted early somatic embryo formation. The suspensor developed by vacuolization and elongation of some cells in early somatic embryo.
