Genetic distance determination between potato (Solanum tuberosum L.) varieties based on Tst1 retrotransposon positions polymorphism

Techniques PCR-IRAP (Inter-Retrotransposon Amplification Polymorphism) and PCR-RBIP (Retrotransposon-Based Insertion Polymorphism) applying Tst1 retrotransposon as DNA marker were used to analyse nematodes resistant and susceptible potato varieties. A tested varieties genetic distance was determined based on an inter-Tst1 copies amplified fragment polymorphism and self inserted Tst1 copies polymorphism. The varieties Europa (RRrr) and Tomensa (Rrrr) have been determined by PCR-IRAP technique as genetically the most distant (DINL 0.818). On the contrary, varieties Europa (RRrr) and Monalisa (rrrr) have been characterized genetically the most similar (DINL 0.273). In the case of PCR-RBIP technique the highest genetic distance (DINL 1.000) has been shown between varieties Monalisa (rrrr) and Fambo (rrrr) and the highest genetic similarity (DINL 0.300) between Europa (RRrr) and Vitesse (RRrr). It is apparent that varieties genetic distance did not reflect the resistance gene presence in the genome. There is a different background of the observed genetic variability based on Tst1 retrotransposon activity. The specific DNA fingerprints, unique by the fragment size and position, have been recorded for individual varieties.
