Cloning and signature analysis of six novel DNA fragments of ribosome inactivating protein from different plants

chinois. 通过分析多种不同来源的植物RIPs的氨基酸序列,据其保守区域设计1对植物通用简并引物P1、P2,对基因组进行PCR扩增,分别从海芋Araceae macrorrhiza,烟草Nicotiana tobaccum、剑麻Agavaccae aga-ve、望江南(Cassia occidentalis、那坡指天蕉Musa spp.(BB)、邻水野蕉Musa spp.(AA)中获得1个基因片段.BLAST分析表明:它们推导的氨基酸序列只与多种不同来源的RIPs有相似性.其中,与葫芦科2个代表RIP相应区段的相似性最高,与β-luffin基因的相似性分别为97.2%、98.5%、97.9%、89.4%、97.9%、97.2%,与天花粉蛋白(trichosanthin,TCS)的相似性分别为63.6%、64.5%、64.1%、60.8%、64.8%、和63.6%.多重序列比对发现:6个序列中分别有10、9、11、11、9、10个残基氨基酸与TCS的N-糖苷酶活性位点的残基完全相同,只是残基位置不一致,仅有个别残基发生了变化.所有的N-糖苷酶活性位点都在P1/P2扩增区段内.
