Rubella antigen (Hem agglutination) was prepared from RA27/3 vaccine strain, by inducing persistent infection n Vero cell line. By this method, mass production of high potent antigen in more simple and economic, because: -Possibility of multiple harvests from a single batch of culture. -Possibility of freezing and storage of persistently infected cells in liquid nitrogen. -Possibility of reviving and re-culture the frozen infected cells for reproduction of antigen. -Possible use of this method for production of rubella vaccine can be considered. By the mentioned method 4000 – 5000 vials of freeze-dried rubella antigen (Hem agglutinin) was prepared, and its potency, specificity and sensitivity were approved by classical methods.
