An extraction method for the recovery of Aphelenchoides besseyi in Oryza sativa seeds by electrolyzed strong acid water

During a study on the application effect of electrolyzed strong acid water (ESW) for the white tip nematode, Aphelenchoides besseyi, we observed that a great number of the nematodes were extracted from Oryza sativa seeds immersed in ESW. Based on the result, investigations were conducted to utilize ESW for accurate estimate of the nematode density in Oryza sativa seeds. We generally followed a mass extraction method of Hoshino and Togashi (2002) for extraction of the nematodes; however, we used a polyester textile instead of a stainless wire net as material of tray for rice seeds to prevent from corroding by ESW, and suspended it in a smaller 300 mL glass container with a 200 mL extracting solution. A diluted ESW containing a pH of 3.5 and 9-10 ppm of available chlorine resulted in greater numbers of the nematodes from dried and 24-hour water-soaked rice seeds (both bisected longitudinally) as compared to tap water. Also, it recovered greater numbers of the nematodes when a re-extraction was conducted immediately after extraction from dried rice seeds with tap water. Adjustment medium solutions (pH 3.5 and/or available chlorine concentration 9-10 ppm) also increased the number of extracted nematodes, but yet the diluted ESW gave highest recoveries of the nematodes. These facts show that ESW is superior in efficiency as extracting solution for recovering Aphelenchoides besseyi from rice seeds to water medium.
