Micropropagated disease-free sweet potato planting material production

Sweet potato is traditionally propagated by stem cuttings. The stem cuttings, however, are exposed to diseases causing microorganisms in the field, which are carried over from one generation to the next causing a reduction in potential yield. Thus, Acedo et al. of the Philippine Root Crops Research and Training Center (PhilRootCrops)-Leyte State University (LSU) [Baybay, Leyte, Philippines] established a micropropagation system wherein clean planting materials can be produced without sacrificing the stability of the morphological and agronomy characters of the plants. To produce a good number of normal stockplants, the culture medium components through additive supplementation. Recommended sweet potato varieties (PSB Sp21, PSB Sp22, PSB Sp23, and NSIC Sp24) were used as test varieties. Findings of the study revealed the following: Grown in modified Murashige and Skoog (MS) medium, the different varieties had 65-84% survival rate with PSB Sp21 having the highest and NSIC Sp24 the lowest. The different varieties had normal meristem growth. To further enhance the production of in vitro stock plants, an experiment to determine the most responsive meristem was conducted. meristems from the different nodal positions in the shoot tissues were used. For PSB Sp23, the best growth was obtained in meristems coming from the fifth node. For PSB Sp22 and NSIC Sp24, the meristems from the third node were more responsive in terms of growth and elongation. The highest percent survival and best growth of in vitro produced plantlets were derived from 1 garden soil (GS): 1 rice hull (RH). Meristem culture technique was considered an important tool in the production of clean sweet potato planting materials.
