Screening of microorganisms for the control of yam anthracnose

Yam (Dioscorea alata L.) is considered as the country's most expensive rootcrop with exemplary nutritional and therapeutic values. It is one of the five banner crops in the Philippines with very high demand in domestic and international market. The tubers however, are very susceptible to anthracnose which is caused by Colletotrichum gloeosporioides. In the Philippine Root Crop Research and Training Center (Philrootcrops) at the Visayas State University, Baybay City, Leyte, the severity of damage of an anthracnose infected area could reach up to 100%. There is a strong need therefore, to control this disease in order to maximize the supply of yam. In this study,a total of 112 endophytes and 94 actinomycetes were screened for anti-microbial activity against C.glocoesporioides using streak and spot inoculation and cell-free supernatant diffusion assay using 8mm paper disc. Endophytes are microorganisms that grow symbiotically inside the plant tissues while actinomycetes are mostly soil microorganisms, which are known to produce antibiotics. This study demonstrated that four endophytic isolates and one actinomycete possess promising antimicrobial activity against the causal microorganism of yam anthracnose-C. gloeosporioides. Three endophytic isolates produce heat-stable compound at 100 deg C, while one isolate produce heat stable compound at 80 deg C. The actonomycetes isolate showed a very promising activity against C. glocoesporioides with zone at inhibition of 36 mm in diameter on agar plate. These isolates are found to be potential source of bioactive compounds that could control yam anthracnose. Further studies are geared towards purification optimization and identification of the compounds produced by these isolates. Potential isolates will likewise be characterized and identified.
