Preimplantational Genetic Diagnosis (DGP) techniques permit to study genetic characteristics of the embryo before the transference of these to the uterus and the implantation takes place. The production of males with the resistant genotype to scrapie disease is contemplated through a MOET program in Rasa aragonesa breed. In the present study we have carry out a technique for sex determination and genotyping of the codons 136, 154 and 171 of PrnP gene in ovine embryos. Sex determination has been have been carry out using of the amelogenine gene and duplex- PCR. To carry out both sex determination and the genotyping of the three codons of the PrnP gene we have used the duplex-PCR, amplifying a repeated fragment specific of Y chromosome and the PrnP gene like an internal control and to carry out the genotyping of three codons of the PrnP gene
