Petrovic, D.
During 2006, it was collected 500 samples of bean lives with symptoms of virus diseases. Out of 500 total samples, 262 samples were collected from 22 different localities. Diseased leaves were divided into 4 groups depending on the type of symptoms (A,B, C, and D). The rest 238 leaves samples were collected from 12 plots in 9 different localities from Vojvodina Province (Serbia) for checking percent of infection in 2006. Bean common mosaic virus, Bean common mosaic necrosis virus, Bean yellow mosaic virus, Cucumber mosaic virus, Alfalfa mosaic virus identification was done by DAS ELISA test. Isolates were proved by test plants. In most plants were present BCMV (35), less was present CMV (8), BCMNV (5%) and AMV (4%), since BYMV was not determined. Mixed viral infection was found in a several samples. The highest percent of infection plants was found in Kovilj (95%), since on Gajdobra and Sremska Mitrovica viruses were not present. In the rest of localities where samples were taken percent of infection range from 10% to 80%. The highest percent of infection founded on plot in Kovilj (Kovilj II - 33%), then in Gospodjinci (13% and 16%), and Kikinda (Kikinda I - 13%), as well non significant infection was determined on the other localities, and range between 1% and 7%. Chosen bean leaf samples were tested using method of reverse transcription and polymerase chain reaction (RT - PCR). Four sets of BCMV specific primers were used for each sample testing. With PCR reaction were proved that BCMV belong to the Potyviridae family, in all three samples was detected BCMV. Set of primers specific for Russian and NL-3D strains were used to prove that determined viruses did not belong to the above mentioned strains.
