Bacterial stalk rot of sweet corn in farmer fields at central growing areas has been studied. A bacterium caused maceration, collapse tissues, slimy wounds with unpleasant smell and stem falling over from Nakhon Ratchasima, Lopburi, Kanchanaburi and Petchabun growing provinces with averaged 8 percent incidence was isolated on nutrient agar (NA). After culturing, the bacterium formed gray-white colonies with circular margin and smooth to rough surface. The bacterium was rod shape, Gram-negative, utilized maltose and cellobiose as carbon sources, and catalase positive. The bacterium could lyse lipid and hydrolyze gelatin but citrate was negative. It grew on potato dextrose agar (PDA) and produced colony types resemble a fried egg. Eighty-four isolates of this causal bacterium exhibited hypersensitive response (HR) on tobacco in 24-48 h. Enzyme plate assay for activity of pectinases and cellulase was determined with 2 selective media contained polygalacturonic acid and carboxymethyl cellulose. Fifty-seven isolates showed clear enzyme activity similar to Erwinia chrysanthemi type strain. Result of three inoculation techniques studied revealed that the dip colony by tooth stick method and sealed with vaseline technique under high humidly could induce 14-days plant suddenly fall over and the lesion enlarged form the base to the stalk rapidly, causing collapse symptom in 1 day after inoculation that showed disease severity level 1. The injection and soil drenching plus root cut wound could induce the same symptom in 3 days after inoculation but they were disease severity level 3. The result suggests that tooth stick tip sealed with vaseline showed highest efficacy of inoculation technique and could be developed to apply for screening bacterial stalk rot resistance in breeding program of sweet corn.
