PCR standardization for detection of babesia ovis in sheep

The polymerse Chain Reaction (PCR) in babesiosis was developed to detect babesia ovis infection in blood sampels containing infacted erythrocytes. two sets of oligonuclotides were designed according to the nucleotide sequences of the 18S rRNA and ATP-binding protein genes of babesia ovis. these primers were used to detect parasite DNA from blood samples of B.ovis-infected blood by PCR. PCR was sensitive enough to detect parasite DNA from 5 mocroliter of blood samples with a parasitemia of 0.000005%. samples from eight isolates of experimentally infected lambs to B.ovis were analysed by using PCR assay.PCR analysis showed that all of the samples were infected by B.ovis. sequence determination and analysis of amplified fragments confirmed specificity of the PCR. the comparsion of two serology and molecular methods in diagnosis of B.ovis infection in experimentally infected lambs showed the PCR detected parasite DNA from earlier stage of infection than did IFAT. these results suggest that the PCR-based diagnostic assay for babesia ovis is highly specific and sensitive.
