Musk of the Ondatra zibethicus Protect Ischemic Injury in the Gerbil Hippocampus

Musk, a dried preputial gland of musk animals, is one of the most famous and expensive chinese medicines. The present study investigated neuroprotective effect of the musk of the Ondatra zibethicus (MOZ) against ischemic insult in the gerbil hippocampal CA1 region after 5 min of transient cerebral ischemia. To elucidate the neuroprotective effect of MOZ on ischemic damage, 50 mg/kg MOZ was administered orally to gerbils for 7 days before the ischemic surgery, and we used cresyl violet, neuronal nuclei protein (NeuN) and Fluoro-Jade C (F-J C) staining. In the MOZ-treated group, many pyramidal neurons in the CA1 region were stained with cresyl violet and NeuN, and a few degenerative pyramidal neurons were stained with F-J C. The survival rate of the CA1 pyramidal neurons was about 60% compared to that in the vehicle-treated group. In the MOZ-treated group, ionized calcium binding adaptor molecule-1 (Iba-1, a marker for microgila) and isolectin B4 (IB4, a marker for activated microglia) immunoreactivity were significantly lower than that in the vehicle-treated group. These results suggest that repeated supplement of MOZ can protect neurons from ischemic damage.
