A research on propagation of Gisela 5 cherry rootstock by tissue culture | Gisela 5 kiraz anacının doku kültürü ile çoğaltılması üzerine bir araştırma

anglais. Gisela 5 was common rootstock in sweet cherry growing in the world. The propagation possibilities of Gisela 5 rootstock was determined by tissue culture in the research. Apical and axilary buds of annual shoots were used as explant materials, and the explants were placed onto MS (Murashige and Skoog) medium containing 1 mg/l IBA (indole butyric acid) + 0.75 mg/l BAP (benzyl amino purine), 1 mg/l IBA + 1 mg/l BAP, 2 mg/l IBA + 0.75 mg/l BAP, and 2 mg/l IBA + 1 mg/l BAP after sterilization of explants. The cultures were incubated in a culture room at 24±2°C with a 16-h photoperiod under 2500 lux light irradiance. The MS basal medium during the rooting stage was supplemented with 0, 1, 2, 4 and 6 mg/l NAA (naftalen acetici acid). The highest shoot number (2.93 number) and shoot lenght (1.68 cm) were obtained from 1 mg/l IBA + 0.75 mg/l BAP and 2 mg/l IBA + 1 mg/l BAP combination, respectively. For rooting stage, different concentrations of NAA (0, 1, 2, 4 and 6 mg/l) were added. 6 mg/l NAA gave the good rooting percentege (92.88 %) during the rooting stage.

turc. Gisela 5, bodur kiraz yetiştiriciliğinde dünyada yaygın olarak kullanılan bir anaçtır. Bu çalışmada Gisela-5 anacının doku kültürü yoluyla çoğaltma olanakları araştırılmıştır. Yıllık sürgünlerin yan ve tepe tomurcukları eksplant olarak kullanılmış ve eksplantlar dezenfekte edildikten sonra çoğaltım aşamasında 1.0 mg/l IBA (indol butirik asit) + 0.75 mg/l BAP (benzil amino pürin), 1.0 mg/l IBA + 1.0 mg/l BAP, 2.0 mg/l IBA + 0.75 mg/l BAP ve 2.0 mg/l IBA + 1.0 mg/l BAP içeren MS (Murashige ve Skoog) besi ortamına alınmışlardır. Çoğaltım ve köklendirme aşamalarında eksplantlar 16 saat aydınlık ve 8 saat karanlık, 24 ± 2°C sıcaklık ve 2500 lux aydınlatma, içeren büyüme odalarına konulmuştur. Köklendirme aşamasında MS ortamına 0, 1, 2, 4 ve 6 mg/l NAA (naftalen asetik asit) ilave edilmiştir. Çoğaltım aşamasında en iyi sürgün sayısı 2.93 adet ile 1.0 mg/l IBA + 0.75 mg/l BAP ve en iyi sürgün boyu 1.68 cm ile 2.0 mg/l IBA + 1.0 mg/l BAP uygulamalarından elde edilmiştir. En iyi köklendirme ise %92.88 ile 6 mg/l NAA uygulamasında saptanmıştır.
