Transformation of wheat high molecular weight gluten subunit14 gene by pollen tube pathway

chinois. 利用优质高分子量麦谷蛋白亚基对小麦进行遗传转化和品质改良。采用花粉管通道法，将小麦高分子量麦谷蛋白14亚基基因导入洛阳8716、陕354、陕893、小偃107和陕150 5种不含该亚基的小麦品种中，转化后代在大田播种出苗后，利用小麦高分子量麦谷蛋白14亚基基因特异性引物进行PCR检测，分析其转化率。不同品种、不同处理间转化率存在很大差异，其中以陕354授粉后切去柱头，滴加50 ng/μL DNA液处理的转化率最高，为1.01％，所有转化材料的平均转化率为0.56％。利用花粉管通道法对小麦进行遗传转化是可行的。_
Afficher plus [+] Moins [-]anglais. The study was to improve the bread-making quality of wheat. High molecular weight glutenin subunit 14 gene was transformed into five wheat varieties by pollen tube pathway. The progenies were grown in field and the leaf genomic DNA of each progenies were extracted. Transformants were identified by PCR using the specific primer of high molecular weight glutenin subunit 14 gene. The result showed that the differences between varieties and treatments were significant, among which the transformation frequency of Shaan 354 with 50 ng/μL DNA was the highest. The average transformation frequency of all varieties was 0.56%. Therefore it is a potential way to transform wheat by pollen tube pathway._
