chinois. 对“杨小-26”南丰蜜桔进行离体培养研究,建立并优化以“杨小-26”南丰蜜桔实生苗为研究对象的无菌培养体系。“杨小-26”种子在MT(Murashige and Tucker)不加任何激素的培养基上培养,长出的实生苗取其茎段、子叶节、上胚轴和下胚轴为外植体分别置于不同培养基上诱导芽。将诱导出的芽转入MT+6-BA0.5+NAA0.05增殖培养基中进行培养,增殖系数可达8.8。MT+NAA0.1培养基有利于‘杨小-26’南丰蜜桔不定根的诱导,生根率可达65.5%。经2d暗培养、6周温室培养之后,移栽于泥炭、珍珠岩、椰子壳(2:1:1)配制的基质中,1个月后幼苗成活率可达90%以上。[著者文摘]
Afficher plus [+] Moins [-]anglais. In vitro culture of Citrus reticulita Blanco ‘Yangxiao 26' was studied. The aseptic micropropagation system of seedlings of Citrus reticulita Blanco ‘Yangxiao 26' was established and optimized. The seeds from ‘Yangxiao 26' were sowed and cultured on MT ( Murashige and Tucker) culture medium without any hormone. The stem segments, cotyledons, epicotyls and hypocotyls chosen from their seedlings were inoculated randomly onto MT culture medium respectively. The shoot multiplication from axillary buds on MT culture medium supplemented with 0.5 mg/L 6 - BA together with 0.05 mg/L NAA could be increased by 8.8 times. Approximately 65.5% of microcuttings rooted on the MT culture medium containing 0.1 mg/L NAA. Over 90% of rooted microcuttings survived in the field, which had been cultured in darkness for 2 days, in glass house for 6 weeks and transplanted in the mixed soil of turf, perlite and putamina of coconut for a month.
