Changes in the blood and lymph nodes of cattle naturally infected by bovine leukemia virus

This work examines blood and lymph node samples from 32 cows, for which the presence of antibodies to the leukemia virus were proven in the blood sera. Full blood was used to make blood smears and a certain number of leukocyte and leukocyte formulas. Samples of retropharingeal, mesenterial, mediastinal, supramammary, and subiliac lymph nodes were fixed in 10% neutral formalin, processed in an automatic tissue processor, and embedded in paraffin. Paraffin sections, around 3-5 micro m thick, were stained with haematoxylin eosin, toluidine blue, and by the immunohistochemical method, streptavidine-biotin (LSAB2) method, using the antibodies for CD3, CD5, CD68, CD79 and vimentin. Microscopic findings were evaluated on the grounds of the criteria used for the examination of lymph node biopsies in humans. In the examined blood samples, leukocytosis was determined in four cows and lymphopenia in two. Other samples (81.25%) had physiological number leukocytes and lymphocytes. An increased percentage and a large number of immature lymphocytes were established in the blood smears of cows with lymphocytosis. In two cows, seropositive to the leukemia virus, a macroscopic examination of the lymph nodes revealed follicular hyperplasia, and diffuse hyperplasia in four cows. Other animals (81.25%) did not have any macroscopic changes. Microscopic examinations of the lymph nodes of cows seropositive to the leukemia virus revealed the following: follicular hyperplasia, T zonal hyperplasia, hyperplasia of marrow cells, mixed type hyperplasia and depletion of lymphatic tissue. CD3+ T lymphocytes are immunodominant population of cells in lymph nodes with t zonal hyperplasia and in lymph nodes with marrow hyperplasia. CD79+ B lymphocytes are predominant cells in lymph nodes with expressed follicular hyperplasia. CD3+ and CD79+ lymphocytes are represent in larger numbers in lymph nodes with expressed mixed type hyperplasia. In lymph nodes with depletion was decrease expression CD3, CD79 and CD68 immunophenotype. Expression CD5 molecule did not revealed in examined lymph nodes.
