anglais. Common blight (Xanthomonas axonopodis pv. phaseoli) (Xap) is one of the most important diseases of bean growing areas in Turkey. In this study, 93 antogonistic bacteria which isolated from healthy bean plants were examined for their ability to reduce bean common blight disease severity. Four different approach were tested for biocontrol effects of the antogonistic bacteria strains against Xap: a) pod inoculation tests b) in vitro tests c) in vivo pot tests and d) field trials. In pod inoculation tests thirty five strains were found effective against Xap by causing 42 to 72; reduction in disease severity in comparison to control treatment. In vitro tests, antagonistic activities of these antogonist bacteria strains were determined according to the both siderophore production and antibiosis. Fourteen effective strains of 35, including fluorescent Pseudomonads and Gram (+) bacteria were also examined for their in vitro plant growth promoting activities. According results, some rhizobacteria strains not only had the ability to produce Indol-3-acetic acid (IAA) at amounts ranging from 2.0 to 33.0 μg/ml, but also had phosphate solubilisation ability. It was also determined that some antogonist bacteria strains increased the root growth by 10 to 60; compared to the non-treated control. In vivo pod tests, all selected strains reduced the diseases severity by 32 to 67 ; compared with the pathogen treatment alone. Phenylalanine Ammonia-Lyase (PAL) activity was determined spectrophotometrically in pathogen inoculated and non inoculated plants which were grown from seeds that were seed-bacterized with five PGPR strains, and inoculated with X. a. pv. phaseoli. The level of PAL in foliar extracts increased significantly upon the PGPR treatment in comparison to the pathogen treatment alone. The reduction in the bean common blight disease incidence was found negatively correlative to the amount of increased level of PAL.

Bu çalışmada fasulye (Phaseolus vulgaris L.) üretim alanlarında görülen önemli hastalıklardan bakteriyel yanıklık etmeni Xanthomonas axonopodis pv. phaseoli (Smith) Dye'ye (Xap) karşı, fasulye bitkisinden izole edilen 93 antagonist bakteri izolatının biyokontrol etkinlikleri araştırılmıştır. İzole edilen antogonist bakterilerin biyokontrol etkinlikleri kapsül inokulasyon testleri, in vitro, in vivo ve tarla denemeleri olmak üzere 4 aşamada gerçekleştirilmiştir. Kapsül testlerinde, 93 antogonist bakteri izolatından 35'i etkili bulunmuş olup hastalık gelişimini kontrol uygulamasına göre % 42-72 oranlarında engellemiştir. In vitro antagonistik etki testlerinde antogonist bakteri izolatlarının siderefor üretimi ve antibiyosis etki mekanizmaları araştırılmıştır. Meyve inokulasyonu testleri ve in vitro biyokontrol etkilerine göre 35 antogonist bakteri izolatından 14'ü etkili bulunmuş ve bu izolatların in vitro bitki gelişimini teşvik edici etkileri araştırılmıştır. In vitro bitki gelişimini teşvik edici etkinin belirlenmesi testlerinde antogonist bakteri izolatlarının 2.0-33.0 μg/ml oranlarında Indol-3-asetik asit (IAA) ürettiği ve bazı izolatların fosfatı çözebilme yeteneğine sahip oldukları belirlenmiştir. Tohum çimlenme testinde ise antogonist bakteri izolatlarının kontrol uygulamasına göre kök gelişimini % 10-60 oranlarında arttırdığı saptanmıştır. In vivo saksı denemelerinde antogonist bakteri izolatları kontrol uygulamasına göre hastalık gelişimini % 32-67 oranlarında engellemiştir. Saksı denemelerinde etkili bulunan 5 antogonist bakteri izolatının bitkide Phenylalanine ammonia-lyase (PAL) aktivitesi üzerine olan etkisi araştırılmıştır.
