chinois. 目的采用real-time PCR方法检测体外激素处理对奶牛乳腺上皮细胞FcRn(neonatal Fc receptor,FcRn)mRNA表达的影响。方法在乳腺上皮细胞培养液中添加不同浓度胰高血糖素(0.01、0.1和1 μmol#8226;L-1)、胰岛素(0.005、0.1和0.5 μmol#8226;L-1)、甲状腺素T4(0.01、0.1和1 μmol#8226;L-1),体外培养并收集细胞,提取细胞mRNA后,利用real-time PCR检测FcRn mRNA的相对丰度。结果在一定添加剂量范围内,随着添加剂量的增加,甲状腺素和胰高血糖素能够显著地促进FcRn mRNA表达量升高(P<0.05),胰岛素在添加量为0.005 μmol#8226;L-1和0.5 μmol#8226;L-1时FcRn mRNA的表达量低,但在0.1 μmol#8226;L-1添加时表达量显著升高(P<0.05)。结论添加外源胰岛素、胰高血糖素和甲状腺素能够影响FcRn mRNA的表达,为进一步研究乳腺内FcRn受体变化以及影响因素提供了依据。
Afficher plus [+] Moins [-]anglais. ObjectiveThe mRNA expression of FcRn was studied in Holstein Cows mammary gland epithelial cells treated with hormone by real-time PCR. Method FcRn mRNA expression of in vitro cultured mammary epithelial cells treated with different concentrations of thyroxine hormone (0.01, 0.1, 1 μmol#8226;L-1), insulin (0.005, 0.1, 0.5 μmol#8226;L-1) and glucagon (0.01, 0.1, 1 μmol#8226;L-1), respectively, was determined.ResultThe results showed that the mRNA abundance of FcRn increased with the higher dose of thyroxine and glucagon significantly (P<0.05) in a certain range. However, the mRNA expression of FcRn was significantly lower when treated with 0.005 μmol#8226;L-1 and 0.5 μmol#8226;L-1 insulin than 0.1μmol#8226;L-1 (P<0.05).ConclusionThyroxine, insulin and glucagon can obviously change the level of FcRn mRNA, which provide a basis for further research of the change and influence of FcRn.
