Expression and analysis of ACE and ACE2 in the kidney of spontaneously hypertensive rats | ACE和ACE2在自发性高血压大鼠肾脏中的表达与作用分析

chinois. 摘 要:为观察血管紧张素转化酶(ACE)和血管紧张素转化酶2(ACE2)在自发性高血压大鼠肾脏中的表达及它们与高血压形成的关系,取14周雄性Wistar-京都种大鼠(WKY)和自发性高血压大鼠(SHR)各8只,自由采食和饮水1周后测定大鼠收缩压(SBP)、舒张压(DBP)和心率(HR),称重后断头处死,取血液和肾脏组织进行如下试验:放免法测定血浆中血管紧张素Ⅱ(AngⅡ)含量,肾脏组织中ACE、AT1、ACE2和Mas mRNA表达水平的Real-time PCR分析,肾脏组织中ACE2蛋白表达的比较分析。试验结果表明:SHR大鼠SBP和DBP均显著高于WKY大鼠,而心率和体重无显著差异(P〉0.05);SHR大鼠血浆中AngⅡ含量和肾脏组织中ACE mRNA表达水平均显著高于WKY大鼠(P〈0.05),肾脏组织中ACE2 mRNA和蛋白表达水平均显著低于WKY大鼠(P〈0.05),2组大鼠肾脏组织中AT1和Mas mRNA表达没有显著差异(P〉0.05)。SHR大鼠肾脏中ACE的mRNA表达显著升高,而ACE2的mRNA和蛋白表达均显著下降。高血压时,肾脏局部组织中ACE和ACE2表达失衡,表现为ACE-AngⅡ-AT1通路的过度活跃,ACE2-Ang(1-7)-Mas通路相对不足。

To observe the expression of angiotensin-converting enzyme(ACE),ACE2 in the kidney of spontaneously hypertensive rats(SHR)and their relationship with hypertension.8 male Wistar-kyoto(WKY)rats and 8 male SHR rats which were about 300 g of weight and 14-weeks old were entrained to free access to food and tap water for one week.Systolic blood pressure(SBP),diastolic blood pressure(DBP)and heart period(HP)of all experimental rats were recorded at the end of experiment,and then all the rats were killed.The plasma and kidney tissue of those rats were immediately collected.The following tests were chosed:the contents of angiotensinⅡ(AngⅡ)in plasma were detected by RIA method;Expression of ACE,AT1,ACE2 and Mas mRNA in heart was analyzed by Real-time PCR method;Expression of ACE2 in kidney was measured by Western blot method.The results showed that SBP,DBP of SHR were obviously higher than those of WKY rats,but there were no significant differences of HR and BW rats in two groups(P0.05);The contents of AngⅡin plasma of SHR rats and the expression of ACE mRNA in kidney of SHR rats increased signifantly compared with WKY rats(P0.05),while the expression of ACE2 mRNA and protein in kidney significantly decreased(P0.05),and the expression of AT1 and Mas mRNA in kidney of SHR rats had a little variance compared with that of WKY rats(P0.05).Compared with WKY rats,the expression of ACE mRNA in kidney increased significantly,while the exprssion of ACE2 mRNA and protein decreased significantly.The expression of ACE and ACE2 in the kindney of hypertensive rats was imbalance which showed that the ACE-AngⅡ-AT1 axis was over-activity and the ACE2-Ang(1-7)-Mas axis was relatively insufficient.
