A large amount of genetic diversity for agro-morphological traits has been showed in annual medics originated from Iran. Inter-intra crossing is one of ways to use this genetic diversity. This study was conducted to give appropriate and applicable method in the country. Seed of annual medics planted in the 15 cm pots were located in greenhouse in October 2006 and 2007. As of smallness of flower, the cross were performed under stereomicroscope. Emasculated and not-emasculated methods used for crossing. A number of 916 flowers were crossed including in 162 intra crossing and the rest inter-crossing. The crossing success ranged from 2,5% for M. rigidula × M. polymorpha to 40% for M. scuttellata × M. turbinate. A high percentage of pods formation after crossing that result to pod death after a while emphasizes on embryo-rescue 2-4 days after crossing and control of the environmental condition to reach a high number of successful crosses. Leaflet shape and plant hairiness can use as early diagnostic traits between parents and offspring. The results of this study showed that the proposed methods can be used for intra- and inter- crossing on originated annual medics from Iran.
