ITALIANO: Com'e' noto gli ioni Al, nei terreni acidi manifestano tossicita' per varie specie, vite compresa. Per questo lavoro si e' studiato l'effetto degli ioni Al sull'attivita' ATPasica della membrana plasmatica, che e' stata isolata da una coltura di calli di vite e purificata per separazione in due fasi acquose. L'Al3+ fu aggiunto ad una soluzione enzimatica a pH 5,5 misurando poi la diminuzione dell'attivita' enzimatica. L'attivita' enzimatica fu ridotta da 1 o 2 mM di Al per piu del 50% nella fase superiore arricchita di ATP-asi. Tale inibizione fu maggiore a pH 5,3. L'attivita' ATP-asica appare ridotta per la formazione di legami fra Al3+ e l'ATP. Aggiungendo acido malico o citrico, che agiscono come chelanti naturali, l'attivita' ATPasica e' ripristinata. Queste conclusioni sono valide per la vite come per altre specie.
Afficher plus [+] Moins [-]anglais. It is well known that Al ions, accumulated in acidic soils, have toxic effect in different plant species. The mechanism of the toxic effect was examined in model experiments. The present work was aimed at the investigation of the effect of Al ions on the plasma membrane ATP-ase activity. Plasma membrane was isolated from callus culture of grapevine and purified by aqueous two phase partition. Al3+ was added into the enzyme assay (pH=5.5) and the reduction of enzyme activity was measured. Enzyme activity could be restored by different chemicals. It can be concluded that the same mechanism works in the case of grapevines as in the case of other plants. ATP-ase activity is reduced by the Al ions because of the binding of Al3+ to ATP. The ATP-ase enzyme activity can be restored by malic acid and citric acid.
