Preliminary studies of bovine leukemia virus in Indonesia.

Bovine leukemia Virus (BLV) is an oncogenic RNA virus, a member of Retroviridae. Since 1969 it has been classified as the aetiologic agent of Enzootic Bovine leukosis (EBL). In Indonesia the situation regarding EBL and BLV infection is not yet clearly recognized, although clinical disease has not been reported. A serological survey was conducted and viral isolation attempted in the present work to study the occurence of BLV infections in cattle in Indomesia. The serological method used was the Agar Immunodiffusion Test using commercial BLV glycoprotein antigen (Pitman-Moore Laboratories). A total of 810 sera collected from West Java, East Java, Lampung and East Kalimantan have been tested. Bali cattle (Bos javanicus), Bos indicus and Friesian Holstein (Bostaurus) cattle were represented. For viral isolation white blood cells of seropositive Bos indicus were inoculated into culture of bovine foetal thymus cells. Whole blood from the infected bovine was also inoculated intravenously into 3 young sheep studied serologically for seroconversion to BLV. 10 of 322 sera from West Java, 6 of 237 sera from East Java and 4 of 129 sera from Lampung reacted in the test. 4 of 10 seropositive animals from West Java become seronegative after 6 months of age, indicated the presence of maternal antibodies. All sera collected from East Kalimantan (122 samples) were negative. A syncytiogenic virus suspected of being BLV was isolated. The 3 young sheep inoculated intravenously with heparinized whole blood seroconverted to BLV within 42 days of inoculation whereas 2 uninoculated control sheep remained sero negative. Confirmation of the isolate as BLV is being attempted.
