Identification of new genes controlling catabolite repression of the synthesis of alcohol oxidase and catalase in the methylotrophic yeast, Pichia pinus.

Mutants of the methylotrophic yeast, Pichia pinus, which are resistant to 2-deoxyglucose during growth on solidified media containing methanol, ethanol, dihydroxyacetone, or succinate, have been isolated. Screening of more than 2000 mutants of this type identified over 300 strains exhibiting constitutive synthesis of alcohol oxidase (AO) in medium containing glucose and approximately 1000 strains with impaired glucose-mediated catabolite repression of this enzyme. In a genetic analysis of these mutants, in addition to gene GCR1 previously identified by the present authors, one new negative type regulatory gene (GCR2) and two new positive type regulatory genes (GCR3 and GCR4) were revealed. These genes control glucose- but not ethanol-induced catabolite repression of the synthesis of alcohol oxidase and catalase. In strains carrying mutations in genes GCR2 and GCR3 (but not GCR4) catabolite repression is impaired and constitutive synthesis of alcohol oxidase and catalase is found in media containing dihydroxyacetone or malate.
