Artificial sporeling culture of seaweed, Gracilaria sp. conducted in RICA Laboratorium was aimed to provide seaweed seeds. The treatment used three methods i.e. aeration, recirculation and stagnant water systems. Nine glass aquaria of 30 x 30 x 30 cm in size and palm-fibre seedling cords of 20 x 20 cm in size were used for this experiment. Sea water used was filtered by 25 micro m filter bag and desinfected with 150 ppt Ca(OCl)2 and neutralized with 45 ppt sodium thiosulfate. Water salinity was decreased until 25 ppt and fertilized with 0.3 ppt TSP and 4 ppt Urea. The results showed that the use of aeration and recirculation systems released spore in shorter time (10-15 minutes) compared with stagnant water system (> 30 minutes). The number of spores released in stagnant water system was more than the other systems. The spore size was relatively not different among three systems i.e. 18-20.4 micro m.
