Cytogenetic analysis of a chromosomal region containing a Drosophila radiosensitivity gene. The effect of juxtacentromeric heterochromatin on mutagenesis of region 44-45 in chromosome 2.

Studies were carried out on the appearance of radiation-induced mutations in region 44-45 of a Drosophila melanogaster chromosome 2 carrying the In(2)G50 inversion, which transfers material from this region to the juxtacentromeric heterochromatin. Fourteen gamma-induced rearrangements with newly-formed break points in region 44-45 of the chromosome containing the inversion were selected. Four of these were deletions, seven were translations, and three were transpositions. The second break point in ten of these aberrations was located in the heterochromatin regions of chromosomes 2 and 3. The frequency of deletion in this region of chromosomes bearing the inversion was found to be an order of magnitude greater than the frequency in the unaltered chromosome, but was nonetheless significantly lower than the frequency observed in similar experiments using other regions of the genome. Thus, a number of features were characteristic for radiation mutagenesis in region 44-45: the predominance of aberrations involving heterochromatin in the spectrum of rearrangements, and the relatively low frequency of deletions; these features are retained when this region is moved to the juxtacentromeric heterochromatin by the inversion.
