Efficacy of bacterial antagonistic formulation for biocontrol of bacterial wilt of tomato caused by Ralstonia solanacearum in greenhouse

The efficacy of antagonistic bacteria formulation (AF) made from Bacillus subtilis strain CH4(BS) amended with different substrates and carriers was evaluated for biocontrol of Ralstonia solanacearum strain To-Ud 3(RS), a causal agent of bacterial wilt of tomato (BWT), under laboratory and greenhouse conditions. Five AFs were developed by mixing BS at 1.8x10sup8 cfu/ml with rice bran and talcum powder (F1), steamed sorghum seeds (F2), F2 + rice bran (F3), sako pellets (F4) and F4 + rice bran (F5). After storage for 3 months at room temperature, F1 formulation had the highest inhibition of RS and increment of BS population (1.6x10sup10 cfu/ml) when tested on NA plate in the laboratory. For biocontrol of BWT in the greenhouse, the F1 formulation provided tomato survival by 66% and tomato yield by 200g/plant whereas non-treated control showed tomato survival by 11% and yield by 10g/plant which were significantly different. The BS population was significantly increased from the initial of 7.3x10sup6 cfu/ml to 5.1x10sup8 cfu/ml while the RS population was significantly reduced from 3.4x10sup8 cfu/ml to 1.24x10sup3 cfu/ml at 8 weeks post inoculation of RS. This study indicated that F1 formulation containing BS: rice bran: talcum powder at 1:2:2 (v/w/w) is effective for controlling BWT until harvesting in greenhouse which is promising for further evaluation in the farmer's field.
