The subject of this biochemical and molecular characterization was the dominant strain ST5 of Klebsiella sp. isolated from oligomineral drinking water from the capped spring in the Vlasina mountaineous region (Serbia). Former data have proven that some types of the genus Klebsiella may be of medical importance as opportunistic pathogens which was the reason why the biochemical and molecular characterization of the strain ST5 had been done. The biochemical characterization of the ST5 strain was performed by means of the manual identification system API 20 E (bioMèrieux). The identification was based on the data base of the identification software (Apilabplus V 3.3.3 bioMèrieux). The molecular characterization was performed by means of the PCR amplification of 16S rDNK and the sequencing method Thermal Cycling Sequencing. The comparison of the NCBI gene bank data for 16S rRNK resulted in the highest level of similarity, i.e. 95% of similarity for the observed genus compared to the genus Klebsiella sp. marked as LB-2 (gi|110931878|gb|DQ831003.1) isolated in China. The biochemical and molecular characterization proved the autochthonous character of the ST5 strain of Klebsiella sp.
