Immunolocalization of 2, 4-D in coconut (Cocos nucifera L.) during sematic embryo genesis

The 2,4-dichlorophenoxyacetic acid (2,4-D) is a small, synthetic and stable molecule that plays a major role in the process of cellular reactivation, acting like a natural auxin. In monocotyledonous plants, small concentrations of 2,4-D allow the proliferation of undifferentiated tissues and its has been widely used to induce somatic embryogenesis. During the process of somatic embryogenesis of coconut, the morphological modifications triggered by 2,4-D. have been well described, however, little is known about the specific site of action of 2,4-D at the cellular level. Using an imaging technique with monoclonal antibodies against 2,4-D. developed by IRD-CIRAD, which have showed high sensibility and specificity and no cross reactions with other endogenous growth regulators in oil palm and date palm, the authors observed this molecule for the first time in coconut tissue and cells during the process of somatic embryogenesis. The visualization was obtained with the use of a fluorochrome linked to a secondary antibody under a confocal laser microscope. In addition, the DAPI dye, specific for double stranded DNA, was used to identify the nuclei. The use of this technique may contribute for a better mastering of the somatic embryogenic process; therefore to improve the efficiency of the propagation of coconut.
