Cloning and sequencing of F protein gene from NR43 Isolate of Iran of Newcastle disease virus

Newcastle disease virus (NDVJ is the type species of the genus paramyxovirus that possese negative-strand RNA genome and causes Newcastle disease in poultry, this virus is important in poultry industry and it causes economicalloss in this industry. Virus and cell membrane fusion mediated by the fusion protein and it is important agent in pathogenicity of virus. For molecular study on ND ~ one of the native isolate from Qom proviance was sequenced. In this research, after extraction of RNA, with .use of specific primers designed for two 800 base pair fragments, we ampilifled these fragments with RT-PCR and PCR products were cloned in T vector and after confirmation with resteriction enzyme digestion, Pstl and EcoRl, two fragments were sequenced. Sequence of first fragment is aligned with NDV sequences in gene bank, and results SIIOW most similarity with Russian strain Vol 95 that we can use this information for designing of recombinant vaccines and other molecular research with regard of native isolated of CO lin try.
