Primary evaluation of seed dormancy in wheat germplasm collection of National Plant Gene Bank

In order to evaluate and characterize the rate of seed dormancy in wheat (which is endemic of Iran) germplasm collection, 1% of germplasm collection were evaluated during a 5 year project. 152 seed samples were randomly selected and cultivated in 5 different sites including; Yazd, Hamedan, Karaj, Kelardasht and Sari, based on a simple systematic design with no replication. Germination test based on ISTA was arranged after both physiological maturity and threshing process in two different temperatures 25°C and 14°C in the darkness. In each experiment 50 seeds were sown in 150 mm petri dishes. Results indicated that: - Seed dormancy exists in wheat germplasm collection varying from 0% to 100%. - Highest seed dormancy was observed in diploids (Agilops) and the lowest seed dormancy was related to tetraploids, the results were the same either after maturity or threshing process. - Based on a randomized block design with 11 genotypes as treatments and three regions as replications, it was observed that, genotypes were significantly different in seed dormancy, and also significant difference between blocks (sites) indicated that genetic characteristics of seed dormancy is affected by environmental conditions. - Statistic results indicated that threshing process and storage are effective factors for breaking primary seed dormancy. - Statistical analysis proved the important role of lower temperature (14°C) in breaking primary seed dormancy.
