Evaluation of effect of high voltage electric fields on mycoplasma in animal sera

Mycoplasma SPP is one of the important contamination factors of vaccines and other biological products wich use of serum.We can eliminate any probably Bacterial contaminations from those products by filtration (except to Mycoplasma ) and Gama radiation. High voltage electric field can destroy the cell membrane and for this reason can kill the cells (such as bacteria); therefore we tried to find a suitable voltage for elimination of Mycoplasma without any destruction on efficiency of sera. At first suitable electrodes and circuit were designed and then the contaminated and uncontaminated sera (with 10 7 CFU/ml of Mycoplasma Agalactiae) were examined by different voltages and number of pulses. After treating all of the sera were tested by growth efficiency test (with 4 different cell culture) and colony counting test (on PPLO agar plate). Finally we could eliminate about 72-75 percent of Mycoplasma Agalactiae by 90 Kv and 30 pulses (0.2 ms) without any decreasing in efficiency of sera. . But in 95 Kv/cm with 35 pulses sera were destroyed and ineffective for cell cultures. Although the sera did not become sterile completely in this attempt but decrease of mycoplasma was significant.Therefore high voltage electric field with these conditions may be helpful for pasteurization of milk and the other pump able foods.
