Environmental isolation of Burkholderia species and seroepidemiology of melioidosis among the indigenous communities in the periphery of Loagan Bunut National Park, Sarawak

The study was to determine the extent of exposure of the indigenous communities to Burkholderia pseudomallei, the causative agent for melioidosis. In addition, an environmental soil survey was conducted for the isolation of B. pseudomallei and related species in the vicinities of Loagan Bunut National Park (LBNP). Soil samples from various ecological sites were screened by culture methods utilising tripticase soy broth (TSB), enrichment threonine-basal salts solution, Ashdown`s media and pseudomallei selective media (BPSA). The reaction profiles kits of the analytical profile index (API GN 32) for bacterial identification system (bioMerieux) was used for biochemical tests on the soil isolates. Single blood samples from the indigenous communities were taken for seroepidemiological studies to determine their exposure to B. pseudomallei by the use of indirect immunofluoresent test (IFA). An environmental soil survey was carried out in various localities demonstrated a high rate of isolation of Burkholderia cepacia from the localities. However, the identification of B. pseudomallei needed to be further clarified by the use of PCR and monoclonal antibodies. The results showed high rate of exposure among the indigenous communities mainly of farmers living in the periphery of LBNP. Melioidosis remains an important occupational health hazard among the indigenous communities.
