Mechanism oxidative damage of cells by t-butyl hydroperoxide and by x-rays

anglais. Cellular DNA can be damaged by direct and indirect influence of ionizing radiation. If the irradiation is carried out by X-rays in the air, approximately 70% of the damage is a consequence of the indirect effect. This means the following: OH-radicals play the main role in DNA damage, because hydrated electrons and H-radicals interact with oxygen. Since the DNA molecules contain bound water, it is possible that OH-radicals are also involved in the direct effect. It was investigate the mechanism(s) of X-ray-mediated cell damage in comparison to mechanism(s) of organic hydroperoxide cytotoxicity and to find the main targets for the two different kinds of cell inactivation. Damage of Chinese hamster fibroblasts induced by tertbutyl hydroperoxide (t-BHP) or X-irradiation was measured by the colony-formation assay and the average single colony volume. Oxidative damage of fibroblasts induced by t-BHP or by X-rays inhibits cell proliferation. The similar decrease of cell proliferation can be caused by DNA dsb without detectable membrane damage (X-radiation) as by membrane damage with nearly no DNA dsb (chemically induced oxidative stress).

russe. Клеточная ДНК может быть повреждена прямым и косвенным воздействием ионизирующего излучения. Если облучение произведено pентгеновскими лучами на воздухе, то приблизительно 70% повреждений является следствием косвенного эффекта. Это означает следующее: основную роль в повреждении ДНК играют ОН-радикалы, потому что гидратированные электроны и H-радикалы взаимодействуют с кислородом. Так как сами молекулы ДНК содержат связанную воду, вероятно,•ОН-радикалы также вовлечены в прямой эффект. Целью нашей работы было выявление механизмов повреждений клеток различными окислительными агентами. Исследовалось содержание SH-групп мембранных белков, накопление субстратов, реагирующих с тиобарбитуровой кислотой, рост колоний и появление двойных разрывов ДНК в культуре клеток В14 китайского хомячка. Повреждение вызывали воздействием рентгеновского облучения, гипохлорной кислотой или трет-бутилгидроперекиси. Полученные результаты указывают на различный механизм окислительных повреждений HOCl в отличие от гидроперекиси или радиоактивности.
