Single nucleotide polymorphisms of the leptin gene and their association to some production traits of sows and piglets (Sus scrofa L.) from different breeds

Obesity in mammals, together with other physical traits, is known to be associated with leptin, a hormone encoded by the LEP gene and is secreted by white adipocytes in response to changes in body weight and energy. By extracting DNA from the hair follicles and employing the PCR-RFLP technique, the associations of LEP gene polymorphisms A1112G and T3469C with body length, back fat thickness, adjusted back fat thickness, average daily gain, weight, and body condition scores of 103 sows, and birth weight, weaning weight, and average daily gain of 153 piglets from a government-accredited breeder farm, were investigated. Very low frequencies of the AA genotype (0.000-0.026) for A1112G and CC genotype (0.000-0.158) for T3469C were computed for all the breeds of sows and piglets. Selection against these genotypes can be inferred. Association of back fat thickness of Large white sows and weight of F1 sows with A1112G were found. Thicker back fat and heavier weight were correlated to AG genotypes. These results may imply selection in favour of Large white sows especially the ones with AG genotype considering that the F sub 1 sows came from Large white and Landrace cross. On the other hand, no association was found between any of the LEP gene genotypes and the traits of piglets. However the piglets of Chester white sows have notably heavier birth weight, and lower weaning weight and average daily gain, especially the AG and TC genotypes, than those of the piglets of Large white sows. Depending on the farm breeding objectives, the results of this study can be beneficial for selection. However, further studies that will use larger number of samples for each breed is highly recommended to obtain more conclusive results.
