Mass propagation of UCB-1 pistachio rootstock through in vitro propagation

The low rate of germination in the seeds of Pistacia species is known as an important problem especially for the seedlings producers. So, micropropagation methods are becoming as a very important tool in pistachio production. In Turkey, disease free and having high multiplication coefficient, facilitating plant quarantine especially in seedlings which come frome abroad to be free from soil diseases and harmful ones make in vitro production of pistachio so important. The aim of this thesis is to investigate the mass production possibilities of UCB-1 rootstock, the hybrid of Pistacia atlantica and Pistacia integerrima in in vitro conditions. For this purpose, the most appropriate applications and environments for the mass production of UCB-1 rootstock were selected by applying different PGR and chemicals related to different surface sterilization which are used for shoot development, budding, and rooting of explants as nodal segments. Sodium hypochlorite (5%, 7%, 10%) was used in surface sterilization and the best sterilization success was obtained by 10 minutes treatment with 10% sodium hypochlorite. To determine the most appropriate shoot development and rooting environments, DKW and Woody Plant Medium as the basic nutrient medium containing Gamborg B5 vitamines and diffrent doses of PGR such as BAP(0, 1, 2, 3 mg/L), IBA (0, 0.1, 0.2, 0.3 mg/L l), NAA(0,0.1,0.2,0.3 mg/L), GA3 (0,05 mg/L), MT (0, 1,2,3 mg/L l) were tested. The best shoot development and maximum shooting were obtained from the combination of 2 mg/L BAP + 0.1 mg/L IBA + 0.05 mg/L GA3. As for the rooting, ½ DKW basic nutrient medium containing Gamborg B5 Vitamins and doses of IBA (1, 2 and 3 mg/L) and NAA '(0.01, 0.1, 1, 0.02, 0.2 and 2 mg/L) was used but root development could not be achieved in these meduim.
