Molecular determination of exopolysaccharide generating abilities of the lactic acid bacteria isolated from traditional cheese

In this study, 800 bp long 16S rRNA's of 39 different lactic acid bacteria isolated from traditional cheese were amplified. The amplified gene regions were sequenced and, with the help of these sequences, microbial identificaton of the species were performed. Besides, the isolates identified through DNA sequences were also defined by electron microscobe. Exopolysaccharide production potentials of the 39 identified isolates were determined by PCR amplification and the amount of the exopolysaccharide they produced was detected by spectrophotometer. Exopolysaccharide production potentials were determined by the amplification of EPSD/E region which has a role in exopolysaccharide generation. With the use of specific primers for the gene region, 200 bp long EPSD/E was amplified in Lb. paracaseiandS.thermophilus strains. Spectrophotometrically, the highest amount of exopolysaccharide generated was measured as 337,19 mg/L, while the lowest one was 57,67 mg/L. The strain with the most capability of exopolysaccharide production was Lb. plantarum, whereas the least production yield was found to be in Enterecoccusspecies.
