Differentiation of potato virus Y strains by N-terminal serology and HPLC peptide profiling

An isolate of potato Y potyvirus (PYV-D) from Delhi, India, was purified and antiserum was produced. In serological tests, unfractionated antiserum reacted strongly with different strains of PYV, eggplant mottle virus and pigweed mosaic virus, suggesting a close relationship between these viruses and PYV. Henbane mosaic potyvirus and cowpea aphid-borne mosaic virus were distantly related. The fractionated antibodies specific to the N-terminal region of the coat protein only reacted with strains ofPYV and eggplant mottle virus but not with pigweed mosaic virus, henbane mosaic potyvirus and cowpea aphid-borne mosaic virus. This suggested differences in the N-terminal regions of the coat protein of these viruses. Reaction with fractionated antibodies specific to the N-terminal region clearly suggested that eggplant mottle is very closely related to PYV. However, the serological tests failed to differentiate strains of PYV. However, peptide profiling of tryptic digests of coat proteins of the strains of PYV by HPLC could clearly differentiate the strains of PYV tested. PYV-D was a distinct strain of PYV.
