Identification of steroidogenic acute regulatory (sTAR) gene in Malaysian indigenous sheep using PCR based substractive hybridization method

A substractive complementary DNA (eDNA) library based on polymerase chain reaction was constructed to identify differential genes in Malaysian Indigenous (Malin) sheep with high twinning rate. Five clones were isolated and sequenced. From the five clones, one clone had an insert that has a 91 % homology with Bos taurus Steroidogenic Acute Regulatory (sTAR) gene. sTAR gene which encoded for Steroidogenic Acute Regulatory (sTAR) protein is essential for adrenal and gonadal steridogenesis. Absence of this gene was shown to cause several impairment to ovarian follicular development and/or ovulation rate of sexually matured 8 weeks old mice. This gene may have influenced the ovulation rate of animals with high twinning rate in Malin sheep population thus causing increased number of litter at a single birth.
