Three flavanones, pinostrobin (l), pinocembrin (2) and alpinetin (3) together with two chalcones, cardamonin (4) and boesenbergin A (5) were isolated from the hexane and chloroform extracts of rhizomes of Boesenbergia rotunda (L.) Mansf. Kulturpfl. The structures of all compounds were elucidated on the basis of spectroscopic methods (IR, MS, and NMR). As a complementary to the spectroscopic data reported previously, the 2-D NMR was carried out to elucidate the structure of boesenbergin A. The extracts and pure compounds were screened for cytotoxic activity against HL-60 (human promyelocytic leukemia cancer cell lines). Cytotoxic screening showed most of the extracts and pure compounds of Boesenbergia rotunda were active against HL-60 cancer cell line. The chloroform extract and boesenbergin A showed the most potent cytotoxic activity with lC50 value of 5.8 µg/ml
