Evaluation of filter paper sero-assay (FIPSA) to detect three viruses

The filter paper sero-assay (FiPSA) was examined, tested and compared with three different plant viruses; Clitoria yellow vein virus (CYW), Hibiscus chlorotic ringspot virus (HCRY) and tomato mosaic virus (ToMY) in purified and crude sap preparations. The filter paper strips that were preblocked were found to increase the sensitivity of FiPSA as the background were suppressed or reduced when compared to strips without pre-blocking in blocking solutions. Positive test was observed as a violet-blue spot. By using pre-blocked paper strips, the purified viruses and sap extracts of CYVV, HCRV and ToMV could be detected up to 3.0 ng/ml and 1: 10,000 dilution, 15.0 ng/ml and 1:4,800 dilution, 5.0 ng/rnl and 1:8,000 dilution, respectively.
