Archaea diversity in tropical coastal waters

The diversity of archaeal communities at Port Klang, Port Dickson, Sungai Muar and Kuala Selangor was investigated with the construction of 16S rDNA gene clones libraries with archaea-specific primers. In total, 1139 randomly selected clones were screened by PCR-restriction fragment length polymorphism (PCR-RFLP) analysis using three different enzymes (Rsal, Cfol, and Ddel). By the values of Shannon diversity (H) index, it was demonstrated that Sungai Muar (H = 3.54) and Kuala Selangor (H = 3.49) clone library had relatively higher diversity of the archaeal operational taxonomic units (OTUs), compared to Port Dickson (H = 2.55) and Port Klang (H = 2.48). Selected phylotypes in this study shared high similarity (≥97%) to Nitrosotalea devanaterra, Nitrosopumilus sp., Methanosaeta concilii, and uncultured archaeon clone sequences. We are currently carrying out further investigations on the effects of different physico-chemical characteristics on the spatial distribution of archaea.
