Effects of different concentration of boron applicated in in vitro conditions on physiological and biochemical characteristics ofsome American rootstocks

This research was carried out to determine the effect of boron on physiological and biochemical characteristics of Kober 5 BB (Vitis berlendierixVitis riparia) and 41 B M.G. (Chaesselas x Vitis berlandieri) American rootstocks. In vitro shoots initiated from mini green cuttings cultured in vitro conditions as plant materials. In this research MS was used as a basal medium and B was added to media at different concentrations of H3BO3 form. These concentrations were 0 (H3BO3 -free), 1.55 mg l-1 (¼ H3BO3 of the standard MS medium), 3.10 mg l-1 (½ H3BO3 of the standard MS medium), 6.20 mg l-1 (standard MS medium), 12.40 mg l-1 ( twofold H3BO3 of the standard MS medium), 24.80 mg l-1 (fourfold H3BO3 of the standard MS medium). The shoots were cultured in the media containing different concentrations of H3BO3 for 2 months.As a result of the study, it was demonstrated that lots of the investigated parameters were affected by the different boron concentrations and the rootstocks. Parameters such as injury degree, shoot length, leaves number, plant weight and rooting affected significantly by the B concentrations. In this study, total chlorophyll varied from 0.43 to 1.18 mg g-1, lipid peroxidation varied from 11.51 to 35.22 nmol g-1 and the amount of hydrogen peroxide varied from 18.43 to 64.60 µmol g-1. On the other hand, vines developed a number of mechanisms such as increasing the amount of proline, soluble protein content and antioxidant enzyme activities including SOD, CAT and APX which take role in removing free radicals to withstand stresses.
