Histology, Histochemistry and Ultrastructure of the Tonsil of Soft Palate in Goats

Histology, histochemistry and ultrastructure of the tonsil of soft palate were studied in six month-old male crossbred goats. The tonsil was located on the nasopharyngeal side of the soft palate as scattered nodules and was lined by pseudostratified ciliated columnar epithelium in the rostral half and stratified squamous epithelium in the caudal part. The epithelium mounting over the lymphoid accumulation was modified into a lympho-epithelium or follicle associated epithelium (FAE). Distinct crypts were not seen. In the lamina propria, only two to three large lymphoid nodules with germinal centers were seen per low power microscopic field. The areas of FAE did not exhibit any PAS reaction. In lamina propria, the fibroblastic reticulum cell (FRC) gave a reticular reaction for acid phosphatase, in the parafollicular and internodular regions. The FRC around the lymphatic nodules and internodular regions showed ATPase activity. Presence of fine-granular ANAE activity was seen in the cytoplasm of T-lymphocytes and macrophages in the FAE, internodular area and mantle zone of the lymphoid nodules. Scanning electron microscopy revealed that the epithelial lining of the FAE of rostral half of the tonsil showed flattened polygonal cells with type I and type II microvillous (MV) cells and a few membranous microfold (M) cells. The caudal part of the tonsil showed many layers of squamous cells with densely packed microplicae. The tonsil of soft palate was not encapsulated.
