Purification and characterisation of novel antibacterial palm kernel expeller (PKE) peptide

Palm kernel expeller (PKE), the most useful by-product from oil palm industry has currently being upgraded into value-added palm kernel product. The alcalasehydrolysed peptide from PKE has proven to have antibacterial activity against sporeforming bacteria. We report herein the characterisation of the purified novel antibacterial PKE peptide. Gel filtration column chromatography was used to fractionate and purify the PKE peptide on the basis of differences in molecular size. The purified peptide, was estimated to have molecular mass around 2.4 kDa as analysed by tris-tricine SDS-PAGE. The purified peptide at 250 μg/mL sufficiently inhibiting the growth of Bacillus cereus. The effect of PKE peptide on the membrane of B. cereus cell was examined by scanning electron microscopy. Our results suggested that, PKE peptide exerts antibacterial efficacy was possibly due to the presence of relavent amino acids, such as leucine and lysine in the peptide molecule and the leakage of the bacterial membrane.
